研究内容:
神经元在特定时间、特定位置、与特定靶标之间形成正确连接,是神经系统能够正确发挥功能的解剖学基础。我们以秀丽线虫为模式生物,结合哺乳动物细胞系,研究神经元与其靶标之间的连接在发育时如何形成,在衰老时如何退行,以及在损伤后如何修复的机制。
研究成果展示
1. P型ATP酶转运体家族P5各成员在神经发育中的作用机制研究:神经元及其连接的发生需要通过神经元细胞膜上受体来接受胞外信号分子的指导。这些胞外信号 (分泌蛋白) 和受体 (跨膜蛋白) 通常在胞质内合成,然后转位到内质网,进入分泌途径或上膜。P5A ATP酶是真核生物P型ATP酶转运体家族的成员,其氨基酸序列从酵母到哺乳动物都十分保守,但P5A ATP酶的底物、转运机制和生理功能都不清楚。我们通过遗传学筛选发现,真核生物中保守存在的P5A ATP酶通过调控神经发育中的关键受体如DMA-1 (Cell Reports 2020, Star Protocols 2021) 和成形素Wnt蛋白转位到内质网的发生过程 (Cell Reports 2021, FEBS Journal 2021, Star Protocols 2022),参与了神经元迁移、树突伸展和轴突导向等发育事件。这些工作不仅发现了P5A ATP酶在神经系统中的生理功能和潜在底物,还揭示了受体和分泌蛋白发生的新调控因子,为进一步研究P5A ATP酶的转运机制打下基础。
2. 非编码RNA对神经元发育和稳态的时空调控:神经连接作为神经元用来接收和传递信息的重要结构,其特异化的形态和结构对于突触传递、信息整合、神经环路形成及可塑性都至关重要。神经连接的发育过程包括了一系列的连续事件,例如神经突的起始、往特定位置的延伸、在靶目标上形成突触等,这些事件触发、衔接和转承的时机都需要精确的调控。我们前期的工作发现两个保守的miRNAs (lin-4和let-7) 在轴突的导向和再生中作为神经元的内在生物钟,起了重要的时序调节作用,介导神经连接复杂而精确的发育过程 (Science Signaling 2012, Science 2013)。最近,我们还通过对双相情感障碍病人的测序发现,mir-27a基因上的一个SNP干扰它的成熟,使得其靶基因NCAM1的水平发生改变,从而影响神经元的迁移和递质释放 (Communications Biology 2022)。这些工作揭示了神经发育和稳态的多重调控机制,为相关神经系统疾病的治疗和药物开发提供了理论基础和潜在靶标。
3. 内质网形态调控蛋白在神经退行性疾病中的作用机制研究:Atlastin (Atl) 作为一类构建内质网形态的类动力蛋白GTP酶,可以促进内质网同源膜的融合。Atls缺失或者突变失活会导致内质网形态异常以及一种神经退行性疾病——痉挛性截瘫 (HSP)。但是Atls是如何被调控来保持内质网动态稳定,以及内质网形态异常和痉挛性截瘫之间存在的关联性都不清楚。我们发现定位于内质网膜上的泛素连接酶Hrd1通过泛素化修饰Atls,帮助维持内质网形态的动态平衡 (iScience 2020)。
